ABSTRACT

Neurological symptomatology is a crucial component of neurological expertise. The committee for fostering board-certified neurologists in the Japanese Society of Neurology (JSN) has hosted an educational activity for residents before the board examination, "The seminar for fostering board-certified neurologists" since 2004. In particular, the autumn seminar has been characterized by small group hands-on of neurological examinations and the evaluations of higher brain function. Besides this, hands-on seminar of neurological examinations by regional branches has been promoted by the JSN. In the Kanto Ko-shin-etsu branch, "The first hands-on seminar of neurological examinations" started in 2023 after a 3-year-suspension due to COVID-19. The most important educational activities in neuromuscular electrodiagnosis (EDx) is the "Neuromuscular diagnostics seminar" hosted since 2004 by the Japanese Society of Clinical Neurophysiology (JSCN). The majority of tutors and participants are neurologists. This seminar is also characterized by small-group, hands-on workshops. We have also run the overseas seminar, "Tokyo Super EMG hands-on," since 2013. These have undoubtedly contributed to enhancing the level of neuromuscular electrodiagnosis not only in Japan but also in other Asian countries. I have conducted studies on neurological symptomatology and neuromuscular electrodiagnosis, which are directly linked to enhancing clinical practice of neurologists through educational activities such as review articles or lectures. Due to the fact that symptomatology is crucial in neurological expertise, neurology is a basic specialty around the world except in Japan. In 2018, the JSN decided to aim to make neurology a basic specialty and continues to make efforts to attain this goal.Copyright © 2023 Japanese Society of Neurology and John Wiley & Sons Australia, Ltd.

ABSTRACT

Based on a participatory study design, this article describes how a group of family members of people deprived of liberty (PDL) experienced the COVID-19 control measures implemented in Mexico's prisons. We conducted 28 in-depth interviews and analyzed them using ATLAS.ti. We found that the measures implemented in Mexican prisons to avoid the spread of COVID-19 focused mainly on suspension of visitation and PDL confinement. The isolation imposed on PDL impacted their living conditions, making them more vulnerable to contracting COVID-19 due to lack of access to essential services, food, and hygiene supplies. Visit restrictions and PDL isolation also impacted PDL relatives' health and socioeconomic conditions. Our findings indicate that the consequences of COVID-19 control actions in Mexican prisons differ according to the gender and jurisdiction of PDL. Women in federal prisons were more isolated, while those in local ones were more deprived of basic supplies. Imprisoned women's isolation has especially severe effects on the mental and physical health of their elderly parents and children. The results show how the measures adopted to control COVID-19 outbreaks in Mexican prisons have exacerbated the preexisting systemic violence experienced by PDL and their families and how they have failed to prevent the spread of COVID-19 in these settings. These findings provide support for the health-informed penal reform of Mexican prisons.

ABSTRACT

Background & Aim: Gene therapies has become recognized for its remarkable clinical benefits in a variety of medical applications, in particular recent approval of an Ad vector-based COVID-19 vaccines have attracted recent global attention. Here, we present key considerations for GMP compliant process development for Coxsackie virus type B3 (CVB3), an oncolytic virus designed for clinical trial in triple-negative breast cancer. Methods, Results & Conclusion(s): CVB3 is a non-enveloped, linear single-strand RNA virus with a size of approximately 27-33 um in diameter. From the initial type using the zonal rotor centrifuge to the advanced type using the tangential flow filtration system and ion chromatograph, we considered the points of the design concept in constructing the manufacturing process. The final design system is constructed as a closed and single-use manufacturing system in which all processes from upstream large-scale cell culture to downstream target purification and concentration steps. In brief, HEK293 cell suspension extended in 3L serum-free medium infected with CVB3, up to 3.6 times 10 to 7 of TCID50 /mL before going to downstream steps, made total 150 mL of final products as 8.43 times 10 to 7 of TCID50/mL concentration. Although further quality control challenges remain that is removal of product-related impurities such as human cellular proteins and residual DNA/RNA to increase virus purity, this concept is effectively applicable even for other types of viruses as GMP manufacturing processes, and would be also important for technology transfer to future commercial production.Copyright © 2023 International Society for Cell & Gene Therapy

ABSTRACT

Aim: A structured education group for adults newly diagnosed with type 2 diabetes has been offered in a face-to- face (F2F) format in the health board since 2009. The suspension of in-person groups due to Covid-19 catalysed redevelopment of the group in a virtual, interactive format. Method(s): The aims and objectives of the virtual group were extended from the original F2F format, and the teaching resources were diversified to include film, animations and a workbook. Patients newly diagnosed with type 2 diabetes, were contacted using a standardised engagement protocol and offered the opportunity to join the virtual group. A series of pilot groups were delivered. The Plan-Do- Study- Act (PDSA) model was used. Each pilot group was studied using mixed method data collection and critiqued by patients, the educator and the team, to improve the delivery methods and patient experience. Result(s): Over six months, eight groups were conducted. Forty-six patients were invited and 30 attended. Engagement was higher in the virtual option compared to usual care prior to the pandemic (65% compared to 55%). Results from feedback forms showed that the majority of respondents either agreed (13%) or strongly agreed (80%) that the group had improved their understanding of type 2 diabetes. The group was given a Net Promoter Score (NPS) of 100. Conclusion(s): The digital option provides a feasible model to deliver an alternative interactive, structured group education programme at diagnosis of type 2 diabetes. The next step involves developing an engagement programme with primary care and application for QISMET accreditation.

ABSTRACT

Objectives: The public's stated preference for public health and social measures (PHSMs), and levels of pandemic fatigue are insufficiently fixed. We aim to quantify the public's preferences for varied PHSMs, and measure population's pandemic fatigue. Method(s): We conducted a cross-sectional, nationwide sampling, survey-based experiment to assess public preference for and attitudes towards PHSMs. A set of psychometric scales, specifically, the COVID-19 pandemic fatigue scale (CPFS), was used to screen fatigue levels in the respondents. The multinomial logit model (MNL) and latent class model (LCM) were utilized for choice tasks analysis, and Mann-Whitney tests were used for CPFS statistical analysis. Result(s): There were 689 respondents, who completed the survey, and were included in the study after quality control. The discrete choice experiments revealed that respondents attached the greatest importance to the risk of COVID-19 infection within three months (45.53%), followed by loss of income within three months (30.69%). Vulnerable populations (lower-income and older respondents) are more sensitive to the risk of infection, and younger respondents are more sensitive to income loss and prefer non-suspension of socialization and transportation. Migrants, and respondents with a higher level of fatigue, have less acceptance of the mandatory booster vaccination and suspension of transportation. Additionally, a higher fatigue level was observed in females, younger respondents, migrants, and relatively lower-income respondents. Conclusion(s): Fatigue and fear of COVID-19 infection contributed to the public's mental health problem. Hence, at the late-stage pandemic, policymakers should consider reducing people's mental burden via relieving people's fear of infection when PHSMs are being relaxed. And this also provides insights for the outbreaks' PHSMs implementation in the future.Copyright © 2023

ABSTRACT

The prevalence of psychiatric disorders namely depression, anxiety, and sleep disturbances has been increased worldwide, particularly during the COVID-19 pandemic. In this regard, the interest of recent investigations is moved toward phytomedicines and bioactive substances derived from natural sources. Although Tilia platyphyllos Scop. contains high amounts of phenolic compounds such as quercetin, kaempferol, and catechin, there is no study on the possible effects of its extract on psychological disorders. The present study was carried out to determine the antidepressant-like, anxiolytic, and sedative-hypnotic effects of the hydroethanolic extract of T. platyphyllos leaves using forced swimming test (FST), tail suspension test (TST), elevated plus maze test (EPMT), pentobarbital-induced loss of righting reflex test and open field test (OFT). Following the ethanolic extraction of T. platyphyllos leaves, the extraction yield was 14% and the total phenolic and total flavonoid contents were found to be 135.23 +/- 0.14 mg gallic acid equivalent/g dry extract and 19.02 +/- 0.03 mg rutin equivalent/g dry extract, respectively. Both FTS and TST revealed a significant antidepressant-like activity for the tested extract at 400 mg/kg compared to the control group. In addition, the anxiolytic activity of the extract was proven through OFT and EPMT in the same dose. Finally, T. platyphyllos extract at 200 mg/kg and 400 mg/kg significantly increased the sleeping time when compared to the control group reflecting its potential hypnotic activity. Co-administration of T. platyphyllos extract at 400 mg/kg and flumazenil as the GABA-A receptor antagonist decreased the sleeping time but the observed effect was not statistically significant. Therefore, we cannot completely rule out the GABA-A receptor's involvement in the hypnotic activity of the extract. The biological results presented here led us to conclude that T. platyphyllos extract can be a prominent source of antidepressant, anxiolytic and hypnotic agents. Probably, the main phenolic compounds of T. platyphyllos such as quercetin, kaempferol, and catechin are involved in the observed effects. ABSTRACT

Commercially available human platelet lysate (hPL) is produced using expired human platelets obtained from accredited blood banks in the United States. These platelets were originally intended for use in patient transfusion. The safety of platelets used in transfusion is managed by the U.S. Food Drug Administration (FDA), as well as the American Association of Blood Banks (AABB). These organizations set standards, including testing for transmissible diseases. The United States record for blood safety is well established, with extremely low rates of disease transmission, making the platelet units used for hPL manufacture low risk. The Covid-19 pandemic has increased awareness of emerging infectious diseases, even though transmission of Covid-19 via blood transfusion has not been documented. For that reason, gamma irradiated hPL offers an additional safety measure in the clinic. Chimeric Antigen Receptor (CAR) expressing T-cells have demonstrated potent clinical efficacy in patients with hematological malignancies. In addition, there are several phase I clinical trials evaluating the use of CAR-T-cells for targeting of solid tumorassociated antigens. Some of the challenging issues found during production of CAR-T cells are the efficiency of T cell transduction to generate CAR-T cells, the expansion of T cells to clinically relevant numbers and the long-term survival in vivo of the therapeutic cells. The use of human platelet lysate has been demonstrated to improve these issues. Our data from experiments performed using human CD3+ from donors demonstrates that human platelet lysates offer an improved performance on T cell expansion versus serum derived products. hPL efficiently promotes T cell expansion, with higher cell yields and lower cell exhaustion rate. Additionally, we efficiently developed a protocol for suspension culture of T cells, which could facilitate the large-scale expansion of allogeneic CAR-T cells.

OBJECTIVE: To assess the willingness of patients with infertility to continue with their in vitro fertilization (IVF) treatment during the COVID-19 pandemic. METHODS: This cross-sectional survey was conducted in the reproductive, endocrine, and infertility medicine department (REIMD) at King Fahad Medical City (KFMC), Riyadh, Saudi Arabia. Patients that were planned to undergo IVF treatment at REIMD were contacted and asked about whether they would like to start IVF treatment during the COVID-19 pandemic from August 2020 to August 2021. Data was analyzed using the SPSS version 24. Statistics obtained as means and standard deviations from continuous variables correlated with the Chi-square test and results were considered significant at p≤0.05. RESULTS: Of the 400 participants, 245 (61.25%) were between the ages of 30-39 years. About 42.75% (n=171) of the patients had 6-10 years of infertility, and 18% (n=72) had at least one pregnancy but no living children. While 64.7% (n=259) of the participants responded on the first call, 83% (n=332) agreed to continue their treatment. Of those, 13% (n=43) preferred to book appointments as soon as possible; 29.8% (n=99) preferred booking within three months; while 57.2% (n=190) chose to book after three months. From our sample, 86.8% (n=59) were afraid to contract the virus and the choice to delay the IVF treatment correlated with the patient's age (p<0.001) and duration of infertility (p=0.007). CONCLUSIONS: The COVID-19 pandemic affected IVF treatment courses, and many patients were afraid to be infected during this pandemic.

The COVID-19 prevention and control measures are taken by China's government, especially traffic restrictions and production suspension, had spillover effects on air quality improvement. These effects differed among cities, but these differences have not been adequately studied. To provide more knowledge, we studied the air quality index (AQI) and five air pollutants (PM2.5, PM10, SO2, NO2, and O3) before and after the COVID-19 outbreak in Shanghai, Wuhan, and Tangshan. The pollution data from two types of monitoring stations (traffic and non-traffic stations) were separately compared and evaluated. We used monitoring data from the traffic stations to study the emission reduction caused by traffic restrictions. Based on monitoring data from the non-traffic stations, we established a difference-in-difference model to study the emission reduction caused by production suspension. The COVID-19 control measures reduced AQI and the concentrations of all pollutants except O3 (which increased greatly), but the magnitude of the changes differed among the three cities. The control measures improved air quality most in Wuhan, followed by Shanghai and then Tangshan. We investigated the reasons for these differences and found that differences in the characteristics of these three types of cities could explain these differences in spillover effects. Understanding these differences could provide some guidance and support for formulating differentiated air pollution control measures in different cities. For example, whole-process emission reduction technology should be adopted in cities with the concentrated distribution of continuous process enterprises, whereas vehicles that use cleaner energy and public transport should be vigorously promoted in cities with high traffic development level.

ABSTRACT

Background: The temporary delay in fertility treatments due to the coronavirus disease 2019 (COVID-19) pandemic, in combination with the imposed lockdowns, has created psychological distress and anxiety amongst infertile patients. Aims: The aim of this study was to evaluate how the pandemic has influenced assisted reproduction technology (ART) patients in Greece, during the second wave of the pandemic. An additional aim was to examine the effects of the pandemic on cross-border patients in particular, compared to national ones. Settings and Design: This study was a cross-sectional, questionnaire-based study, distributed to 409 patients of a single in vitro fertilisation (IVF) clinic in Greece, during the period between January until the end of April 2021. Materials and Methods: The survey was conducted online via E-mail and was distributed to national and international female patients of a single IVF clinic in Greece, who were undergoing ART treatment during the second wave of the COVID-19 pandemic. Patient participation was anonymous, and participants provided informed consent for collection and publication of data. Statistical Analysis Used: The mean values of baseline characteristics, along with answer percentages per questionnaire item, were calculated. Collected data were cross-tabulated, and the Chi-square test was used as a measurement of the differences between national and cross-border patients. A P value lower than 0.05 was considered statistically significant. All analyses were conducted using the SPSS Statistics software. Results: From 409 initial candidates, 106 women, with a mean age of 41.2 years, completed the questionnaire (26% response rate). The majority of national patients did not experience any delays in their fertility plans (62%), while cross-border patients experienced over 6 months of delays (54.7%). The main reason for fertility postponement was travel restrictions due to COVID-19 for cross-border patients (62.5%), while national patients cited additional reasons. The majority of patients experienced a degree of stress (65.2%) due to the delays, however were not fearful of COVID-19 infection (54.7%). Most patients were aware of the protective measures taken by IVF clinics (80.2%), and this was a determinant factor (71.7%) for their decision to restart their fertility treatment. Conclusion: The COVID-19 pandemic lockdowns had a significant emotional impact on patient receiving or undergoing ART treatment in Greece. This impact was more pronounced on cross-border patients. This highlights the need for continuation of ART care, with the appropriate protective measures, during the pandemic, as well as during similar times of crisis in the future.

Suspension and expulsion have been documented concerns in childcare centers throughout the United States for nearly 20 years. This study examined suspension and expulsion practices in community childcare centers two years into the COVID-19 pandemic (May 2022). Survey data from 131 administrators of community childcare programs were analyzed. It was found that at least 67 individual children were reported expelled across 131 programs, a rate similar to pre-pandemic rates and higher than rates at the height of the pandemic. For suspension, 136 individual children were suspended from early learning programs during this time; a rate almost double pre-pandemic levels. Factors (availability of support, prior suspensions, suggesting the program is not a match, reported turnover, waiting lists, enrollment capacity, administrator reported stress, and teacher perceived stress) were examined to determine if they predicted expulsion. None of these factors significantly predicted expulsion. These results and their limitations and implications are discussed.

Blood clotting has become one of the most dangerous side effects associated with Corona virus, as well as the high level of cholesterol and triglycerides in the blood. Therefore, it has become necessary to use medicinal plants that are biologically safe and containing anti-clotting compound. Feijoa sellowiana represents a prolific source diverse compounds that may have thrombolytic activity. Therefore, the main research point is the production and scaling up of a target contents that have anticoagulants by using biotechnological techniques; calli production, and bioreactors and assessed their activity through in-vivo study. Murashige and Skoog (MS) medium enriched with varying concentrations of benzyl adenine (BA) and naphthalene acetic acid (NAA) was used to cultivate calli and cell suspension cultures from F. sellowiana seeds. Bioreactors were employed to boost active constituent's production. Moreover, the bioreactor physical factors such as effect of controlled or uncontrolled pH medium were investigated. The leaves of the main plant were extracted by ethanol 70% and polar and non-polar extracts were also prepared. The ethanol extract of calli and cells resulting from bioreactors were also prepared. All prepared extracts were subjected to chemical analysis by HPLC, in-vitro antioxidant assays, in-vivo anticoagulant activity and histopathological examination. Calli and cell suspension cultures were produced by using MS medium fortified with 1 mg/L BA+ 0.1 mg/L NAA. It was found that culturing of cell cultures in a bioreactor with uncontrolled pH and aeration at the value of 0.5 L/min gave the maximum and economical fresh and dry weights of the plants. After evaluation of all extracts; it was found that the calli ethanol extract for each plant was the highest value of total phenolic and total flavonoid contents either quantitatively or qualitatively. All extracts of Feijoa had antioxidant activity. The IC50 of the DPPH of Feijoa calli extract was 13.45 µg/mL, it was also confirmed by FRAP and ABTs values. Feijoa calli extract decreased platelet aggregation by suppression of thrombin, extended aPTT, PT, bleeding and clotting times. It was safer than warfarin medication. From these findings the authors can conclude that Feijoa had highly anticoagulant activity and the calli production achieved the goal of the enhancement of the phenolic constituent and thus their activity.

Background/Aims There are no data on the collective incidence of the large vessel vasculitides. The incidence of GCA and Takayasu arteritis in the UK has been based on clinical coding in routine administrative datasets. There are no data on the incidence of these diseases based on clinically verfied diagnoses. We studied the incidence of the large vessel vasculitides in a stable population with a predominant Northern European ancestry. Methods Individuals attending a secondary care hospital with a clinically verified diagnosis of primary systemic vasculitis made between 2011-2020, who lived within the NR postcode districts of Norfolk county were included if they met classification criteria for GCA or Takayasu arteritis, or had definite tissue or imaging evidence of large vessel vasculitis. Population above the age of 18 as in the 2011 census, available from the office of national statistics, was accepted as the denominator. If classification criteria for both GCA and TAK were met, physician judgement was accepted as final diagnosis. Results 272 adults were diagnosed with large vessel vasculitis out of a population of 454,316. The annual incidence of large vessel vasculitis in Norfolk is 59.9/million in the adult population. The annual incidence of GCA is 9.9/100,000 over the age of 50 using the ACR 1990 criteria and 10.6/100,000 using the ACR/EULAR 2022 criteria. The rise in the incidence from 2017 onwards coincides with the establishment of a fast-track pathway (Table 1). The dip in the incidence in 2020 coincides with suspension of services during the SARS-CoV-2 pandemic. The annual incidence peaks at 168.5/100,000 in the 9th decade and is commoner in females (12.3/100,000 vs 7.3/100,000). The annual incidence of Takayasu arteritis is 3.3/million in the adult population using the ACR 1990 criteria and 1.1/million using the ACR/EULAR 2022 criteria. Conclusion This is the first study that reports the incidence of all objectively diagnosed large vessel vasculitis in a stable population in Norfolk county. The incidence of GCA rose with the establishment of a fasttrack pathway and its peak may have been affected by the SARS-CoV- 2 pandemic. GCA is commoner in females and peaks in the 8th and 9th decades. (Table Presented).

ABSTRACT

The present study narrates the extraction of rutin from Tagetes erecta (Marigold) via maceration followed by ultrasonication. The extracted rutin was further fabricated into nanoparticles by high-pressure homogenization (HPH) and assessed by HPLC, DSC, XRD, TEM, and FTIR spectroscopy. The optimized batch of nanoparticles obtained using 32 central composite design (CCD) which exhibited particle size 209±14 nm, PDI 0.234±0.06, and 92±1.3% entrapment efficiency. The lyophilized rutin nanoparticles were further converted into nano-suspension. Interestingly, the rutin nano-suspension exhibited a similar antitussive effect in vivo as that by standard treatment pentoxyverine and reduced the coughing times within 2 min. Also, the phlegm showed high UV absorbance, implying its better expectorant activity than the standard and control. The rutin nano-suspension was highly stable and shelf life was found to be ∼29.1 months. The present study, for the first time, paves a way for the use of rutin nano-suspension to overcome chest congestion, shortening of breath, and in the management of cough.

Background and Aims: The covid-19 pandemic has reversed years of progress in the fight to end tuberculosis. So, the discovery of new drugs as antituberculosis is very much needed. Our previous studies have shown that the extract of Centella asiatica is able to inhibit the growth of Mycobacterium tuberculosis in vitro and requires further research. The aims of this study is to prove the effect of Centella asiatica inhibit Mycobacterium tuberculosis in rat model tuberculosis. Method(s): The protocol in this study was approved by the veterinary ethics committee of Airlangga University. The rat tuberculosis model was induced by intrathecal injection of a suspension of Mycobacterium tuberculosis strain H37 Rv. Twenty-eight tuberculosis rat were randomly divided into four groups. Groups 1,2, and 3 were treated with ethanol extract of Centella asiatica at 375 mg/kgBW, 750 mg/kgBW and 1500 mg/kgBW, and the fourth group was the control group. Centella asiatica extract is administered orally via an intragastric feeding tube for two weeks, once daily At the end of the experimental period, rats were sacrificed by cervical decapitation. The left lung tissue was taken aseptically and cultured on Middlebrook 7H10. Result(s): The results showed that there was no bacterial growth on the culture media in the group that received Centella asiatica extract at a dose of 750 and 1500 mg/kg BW. ABSTRACT

Background: Retrospectively quantifying effectiveness of interventions such as travel restrictions to counter viral introduction and transmission is critical to inform public health policy. Phylogenetic analyses of SARS-CoV-2 variants were undertaken to quantify the effects Canadian COVID-19 travel restrictions had on variant importation and transmission dynamics. Method(s): Global and Canadian GISAID sequences available up to March 2022 were subsampled proportionally to variant-specific case counts and ten phylogenies were inferred for each variant. Trees, dates, and geographies were inferred using maximum likelihood. Result(s): In response to Alpha, Canada implemented a UK flight ban from December 20, 2020-January 6, 2021, resulting in a 1.5-fold reduction in UK sublineage importation rate, with subsequent rebound (Fig. 1). Enhanced screening measures were implemented on December 24, 2020 for South African arrivals to counter Beta. Although there was a 6.3-fold reduction of Beta sublineages per week from South Africa following enhanced screening, there is low confidence in rare events. For Gamma, enhanced screening for arrivals from Brazil was implemented March 31-April 13, 2021. Proportion of Gamma sublineages from Brazil was reduced 1.6-fold within 2 weeks of the intervention, but the weekly importation rate was not significantly changed from start to end of intervention. In response to Delta, Canada issued a suspension of flights from India from April 22-September 23, 202, coinciding with a 2.4-fold reduction in sublineage importation and 3.8-fold reduction in proportion of sublineages from India. Increased importations from the USA and Europe progressively negated the ban's effectiveness. Against Omicron, Canada banned entry of all foreign nationals who had travelled through southern Africa and implemented enhanced screening for Canadians from November 26- December 18, 2021. Subsequently, the BA.1 sublineage importation rate from South Africa was maintained at a low level amid rising cases, while importations from other sources increased, reducing the proportion of sublineages from South Africa and diluting the measure's effectiveness. Conclusion(s): Flight bans and enhanced screening against SARS-CoV-2 variants were most effective when implemented rapidly and for lengthier time;however, effectiveness declined as variants became globally widespread. Ongoing genomic surveillance programs incorporating phylodynamic analyses can inform travel restriction and non-pharmaceutical intervention policy. (Figure Presented).

ABSTRACT

School suspension and expulsion are not limited to middle and high school students;these exclusionary approaches to discipline begin in early childhood settings such as elementary schools. Most studies regarding exclusionary discipline have been conducted on the middle and high school level, but there are very limited studies regarding exclusionary discipline in early childhood or elementary school level. African American elementary school students are disproportionately represented in multiple school suspensions. Although the majority of school suspensions were not due to violent school crimes, but for discretionary offenses, zero tolerance policies have substantially contributed to students being suspended or expelled for trivial infractions. The purpose of this study was to use qualitative research to investigate New York City's public elementary school teachers' perceptions of African American students' behaviors, and how teachers would describe effective behavior intervention approaches for African American students. Data were collected through the use of interviews and journal prompts completed by teachers during the fall of 2021. Results from this study showed that African American students were perceived by the teacher participants as having similar behaviors to other children of various racial and ethnic groups, and needed effective behavior intervention approaches that addressed mental health counseling, extra-curricular and movement activities, preventative measures to discipline, and culturally responsive education. The results obtained from this study may inform educators and policymakers of social justice alternatives to exclusionary discipline. Further studies are needed to examine the "sit still culture" and the role of movement activities and kinesthetic learning modalities as behavioral intervention approaches to lessen disruptive behaviors and curb punitive exclusionary practices, as well as the effects of social justice alternatives to exclusionary discipline as a support system for black students post COVID-19. ABSTRACT

The human cell line HEK293 is one of the preferred choices for manufacturing therapeutic proteins and viral vectors for human applications. Despite its increased use, it is still considered in disadvantage in production aspects compared to cell lines such as the CHO cell line. We provide here a simple workflow for the rapid generation of stably transfected HEK293 cells expressing an engineered variant of the SARS-CoV-2 Receptor Binding Domain (RBD) carrying a coupling domain for linkage to VLPs through a bacterial transpeptidase-sortase (SrtA). To generate stable suspension cells expressing the RBD-SrtA, a single two plasmids transfection was performed, with hygromycin selection. The suspension HEK293 were grown in adherent conditions, with 20% FBS supplementation. These transfection conditions increased cell survival, allowing the selection of stable cell pools, which was otherwise not possible with standard procedures in suspension. Six pools were isolated, expanded and successfully re-adapted to suspension with a gradual increase of serum-free media and agitation. The complete process lasted four weeks. Stable expression with viability over 98% was verified for over two months in culture, with cell passages every 4-5 days. With process intensification, RBD-SrtA yields reached 6.4 µg/mL and 13.4 µg/mL in fed-batch and perfusion-like cultures, respectively. RBD-SrtA was further produced in fed-batch stirred tank 1L-bioreactors, reaching 10-fold higher yields than perfusion flasks. The trimeric antigen displayed the conformational structure and functionality expected. This work provides a series of steps for stable cell pool development using suspension HEK293 cells aimed at the scalable production of recombinant proteins.

We explored whether coronavirus disease 2019 (COVID-19)-related distress was associated with voluntary suspension of medically-assisted reproduction (MAR) treatment from April to May 2020 in Japan. Data for 1,096 candidate respondents were collected from a Japanese nationwide cross-sectional internet survey distributed from August 25 to September 30, 2020. Multiple logistic regression was performed to clarify the association between voluntary suspension of MAR treatment and the Fear of COVID-19 Scale (FVC-19S) score. High FCV-19S score was inversely associated with voluntary suspension of MAR treatment compared to low FCV-19S score among women (OR = 0.28, 95% CI = 0.10-0.84). Age-stratified analyses revealed that low FVC-19S score was significantly associated with voluntary suspension of MAR treatment among women aged < 35 years (OR = 3.86, 95% CI = 1.35-11.0). In contrast, the association between FVC-19S score and voluntary suspension of MAR treatment was reversed and not significant among women aged ≥ 35 years (OR = 0.67, 95% CI = 0.24-1.84). COVID-19-related distress was significantly associated with voluntary suspension of MAR treatment among women aged < 35 years, and this association was reversed but not significant among women aged ≥ 35 years.
